Summary

Xenotransplantation is a potential solution to addressing the global shortage of transplantable human tissues and organs. The most significant hurdle to clinical xenotransplantation is immunological rejection of the transplanted xenograft. Components of the innate immune system have been known to have an enormous influence in the rejection response. This thesis investigates several rejection mechanisms of pancreatic islet xenografts, with emphasis on preformed antibodies against the galactose-a1,3-galactose (aGal) carbohydrate, the potential roles of innate responding cells such as macrophages, inflammatory cytokines and chemokines, as well as pattern-recognition receptors like the toll like receptors (TLRs).
